要約(日本語): This work demonstrates the photoinjection of DNA plasmids into Tobacco BY-2 cells by a small pore formation using single femtosecond laser pulse. The injection process was visualized by utilizing fluorescence-labeled DNA plasmid. The injection was investigated with osmotic pressure control and pH control of culture medium. The osmotic pressure control was utilized to reduce the turgor pressure. Decreasing pH of culture medium reduces the interaction between DNA plasmid and the cell wall and softens the cell wall. By optimizing these effects due to the osmotic pressure and pH conditions, DNA plasmids could be injected into intact Tobacco BY-2 cell.
