要約(日本語): Immunosensor has high sensitivity and specificity of antibody. In this study antigen-antibody binding is detected by fluorescence anisotropy change of fluorescently labeled antibody. With binding of antigen, molecular motion of antibody Fab region is restricted and fluorescence anisotropy increases. This sensing scheme is theoretically free from the background noise of surface adsorption. A evanescent excitation optical stage was constructed for antibody immobilized substrate measurement. Feasibility of this method was investigated by the measurements of fluorescence anisotropy of sensor substrate by evanescent excitation system. Anisotropy of immobilized antibody was changed by administration of antigen molecule. Sensitivity of the system was lower than 1 pg/mL.
