要約(日本語): We had previously proposed a novel odor biosensor by expressing ionotropic odorant receptors of insects into dissociated cultures of neurons of rats. The odorant-gated ion channel structure of insect odorant receptor could realize easy functional expression into cells. The neuronal dissociated cultures of rats have three significant advantages: a long lifetime comparable to rats, i.e., a few years; amplification ability from weak ionic currents of odorant receptors into easily detectable action potentials of neurons. In the present work, we investigated whether dissociated cultures with transected cells could change the spatio-temporal activities in response to different odor stimuli. We demonstrate that cultures with 10% of pheromone-receptor-expressed cells showed synchronous increase of calcium signals at the presentation of pheromone in both transfected cells and non-transfected cells and that the activity pattern was dose-dependent. Thus, these results demonstrated that, in our proposed senseor, discrimination of odorants from spatiotemporal activity pattern is possible.
